For PCR fluorescence analysis, the LED light source used in real-time fluorescence PCR (qPCR) instruments must possess high stability, narrow spectrum, high coupling efficiency, and low drift characteristics to ensure the excitation efficiency and detection sensitivity of fluorescent dyes (such as FAM, HEX, ROX, Cy5, etc.).
Regarding LED wavelength selection, the following light sources are prioritized to match mainstream fluorescence channels:
430nm peak wavelength LED (suitable for FAM, SYBR Green excitation)
470nm peak wavelength LED (suitable for FAM/FITC excitation)
520nm peak wavelength LED (suitable for HEX/VIC excitation)
4000K full-spectrum white LED (for broadband excitation or auxiliary illumination)
630nm peak wavelength LED / 3000K white LED (suitable for ROX/Texas Red excitation)
740nm peak wavelength LED (suitable for Cy5/Cy5.5 excitation)
